چکیده
Purpose Conjunctival impression cytology is a reliable method to obtain epithelial cells from the ocular surface. Its application has broadened our knowledge of surface disease, paving way for greater understanding pathogenesis new treatment options and personalised medicine. Herein we compare most commonly used materials optimise isolating microRNA (miR) conjunctiva. Methods MiR yield 12 mm PTFE 13 PES in free membrane semicircle form were compared (n = 3). Next single Eyeprim applicator 8). MiRs isolated using miRNeasy kit (QIAGEN GmbH Hilden Germany) concentration was determined on Nanodrop ND-1000 Spectrophotometer (Thermo Fisher Scientific Wilmington DE). In second experiment, subjective discomfort provoked by each technique also SANDE immediately 2 min after sampling. Results had significantly higher miR vs PTFE; 131.3 ng/µl±8.1 32 ng/µl±9.9 ng/µl p 0.0002. experiment comparable Eyeprim; 48.7 ng/µl±24.4 46.9 ng/µl±19.2, 0.88. measured participant this experiment. Regarding intensity tolerable at point sampling; 0.1± 0.3 4.5±2.8 Eyeprim, 0.0016. At min, 0.1 ± 3.3 3.22 0.03. Conclusions Our results confirm that greatly superior its harvesting ability which previously optimised studies Millicell Biopore. Furthermore, commercially designed while being better tolerated. We recommend use 0.2 µm pore design harvest characterise expression conjunctival cells. This simple, inexpensive can easily be implemented clinical research setting investigate disease.

Impression cytology of the ocular surface.
Impression cytology refers to the application of a cellulose acetate filter to the ocular surface to remove the superficial layers of the ocular surface epithelium. These cells can then be subjected to histological, immunohistological, or molecular analysis. Proper technique is essential as the number of cells sampled can vary considerably. Generally two to three layers of cells are removed in ...
متن کاملReliability of impression cytology for the diagnosis of ocular surface squamous neoplasia employing the Biopore membrane.
AIM To evaluate the accuracy of impression cytology employing a Biopore membrane device in the diagnosis of ocular surface squamous neoplasia (OSSN). METHODS The histology of patients undergoing excision biopsy for the suspected diagnosis of OSSN over a 20 month period was compared with the reported cytology of impression cytology specimens obtained preoperatively using the Biopore membrane d...
متن کاملImpression cytology and ocular characteristics in ocular rosacea.
PURPOSE To examine clinical findings and histologic changes on the conjunctival surface in ocular rosacea. METHODS Thirty-five patients with ocular rosacea and 30 normal subjects underwent dermatologic and ocular examinations. Tear film break-up time, Schirmer tests with and without topical anesthesia, and conjunctival impression cytology were done. Patients were divided into the following gr...

Purpose: To evaluate the agreement between the methodologies of impression cytology (IC) and histopathology regarding epithelial lesions clinically diagnosed as pterygium and also regarding the detection of unsuspected and associated ocular surface squamous neoplasia (OSSN). Methods: Thirty-two Brazilian patients were included and IC was performed on all pterygia before excision. Histopathogica...
متن کاملImpression cytology following mitomycin C therapy for ocular surface squamous neoplasia.
BACKGROUND/AIMS Topical mitomycin C (MMC) therapy has been used for treatment of ocular surface squamous neoplasia (OSSN) since 1994. Relatively few studies have reported the cellular changes in ocular surface following MMC. METHODS Impression cytology was studied in four patients with ocular surface squamous neoplasia, either primary or recurrence after previous excisional biopsy. The author...
